Matsuo began his career as a child actor at the Shinpa Theater in 1920, at the age of four. He made his film debut in 1924 and worked for several studios, including Makino Film Productions and Kawai Productions, playing lead roles in silent films.
He joined Toho in 1941 and transitioned into supporting roles after the war, appearing in numerous films such as Rodan (1956) and The Human Vapor (1960). He also had roles in the Samurai trilogy alongside Mifune Toshiro.
After leaving Toho in 1959, he continued to work as a freelance actor, primarily in television dramas, into the 1960s.
