Murakami graduated from Tokyo Imperial University (now the University of Tokyo) with a degree in Economics. He began his acting career after serving in World War II, joining Toho in 1950. With his intellectual demeanor, he became the studio's go-to actor for roles requiring gravity and intelligence, such as scientists, doctors, and government officials.

He is best known to genre fans for his role as Dr. Tanabe in the original Godzilla (1954). He also played the mad scientist in The Human Vapor (1960) and a government minister in Invasion of Astro-Monster (1965). In 1968, he co-founded the Romant Theater company, where he mentored younger actors. He continued to act until his death in 2007.
